ABSTRACT

As raças taurinas de origem ibérica Limonero e Carora (Bos primigenius taurus) possuem o fenótipo de pelo curto, liso e com baixa densidade folicular, o que confere a esses animais maior tolerância térmica e melhor produtividade em regiões quentes. Diferentes mutações associadas a esse fenótipo foram descritas no gene do receptor de prolactina PRLR, localizado no cromossomo bovino BTA20. Uma mutação recentemente encontrada é a substituição do nucleotídeo C por T, SNP 39136666 (p. R497*), no exon 11, que gera um códon de parada e, consequentemente, uma menor isoforma desse receptor. Neste trabalho, desenvolveu-se um protocolo rápido e de baixo custo para detecção desse SNP, utilizando-se a técnica de tetra-primer ARMS-PCR. Assim, foi possível detectar essa mutação nas raças brasileiras de origem ibérica localmente adaptadas: Caracu, Crioulo Lageano, Mocho Nacional e Pantaneiro. O alelo T foi mais frequente na raça Caracu (80%), enquanto o alelo C foi mais frequente na raça Crioulo Lageano (84%). Essa simples metodologia pode ser usada para genotipar esse SNP e ajudar na aplicação dessas informações moleculares em programas de melhoramento focados na tolerância térmica em bovinos taurinos e seus mestiços.(AU)

ABSTRACT

Embryos of Caiman yacare were collected and subjected to the bone clearing and staining protocol in orderto analyze the ontogenetic patterns of ossification of the pectoral girdle and forelimb skeleton. The osseousstructure of the girdle and forelimbs of C. yacare begins to ossify starting at 30 days of incubation, withthe presence of dye retention in the scapula, coracoids, humerus, radius and ulna bones. During embryonicdevelopment, the autopodio of C. yacare has four bones in the carpus, the radial, ulnar, pisiform and carpaldistal 4+5 bone. Their ossification begins at 39 days of incubation with the radial, followed by the ulnar, and at54 days, the pisiform and the distal carpal 4 + 5. Each mesopodio has 5 metacarpi and are present 15 phalanges,two in digits I and V, three in digits II and IV, and four in digit III (phalangeal formula 2:3:4:3:2). Ossificationof the metacarpi starts at 27 days of incubation, following the sequence MCII=MCIII=MCIV>MCI>MCV.The first phalanges begin the process of ossification on day 36, continuing up to the last day of incubation.The sequence of ossification of the proximal phalanges is PPI=PPII=PPIII>PPIV=PPV, that of the medialphalanges is MPII>MPpIII>MPdIII>MPIV, and that of the distal phalanges is DPI>DPII>DPIII>DPV>DPIV.The ontogenetic pattern of the bones of the forepaw of C. yacare generally differs from that of other reptiles,although there are some similarities.

ABSTRACT

Hematological and biochemical parameters, including plasma electrolytes and thyroid hormones, were determined in 73 clinically healthy Churra-da-Terra-Quente ewes, a typical breed from the northeast of Portugal. The hemogram values were: erythrocytes 9.8±1.5×10(12)/L; haemoglobin 118.1±19.1g/L; haematocrit 40.8±5.9 percent; leukocytes 5.7±1.8×10(9)/L; and platelets 544.3±177.2×10(9)/L. The thrombin time was 17.3±1.7 seconds. The values of biochemical parameters were: total protein 76.4±6.1g/L; glucose 2.87±0.60mmol/L; total cholesterol 1.65±0.33mmol/L; aspartate aminotransferase 155.9±49.2U/L; alanine aminotransferase 23.2±9.6U/L; γ-glutamyl transferase 48.0±18.7U/L; total alkaline phosphatase 121.6±76.1U/L; glutamate dehydrogenase 6.4±3.7U/L; urea 7.32±2.22mmol/L; creatinine 123.0±54.1μmol/L; total calcium 2.53±0.25mmol/L; phosphorus 2.10±0.46mmol/L; magnesium 1.01±0.09mmol/L; sodium 152.04±3.65mmol/L; potassium 4.7±0.4mmol/L; ionized calcium 1.32±0.07mmol/L; total thyroxine 111.75±42.29nmol/L; total triiodothyronine 1.01±0.28nmol/L; free T4 11.93±1.78pmol/L; free T3 4.22±1.33pmol/L; and thyroid-stimulating hormone 0.18±0.19μIU/mL. Although differences among the Churra-da-Terra-Quente breed and other breeds may occur, the hematological and biochemical parameters, plasma electrolytes, and thyroid hormones, for this indigenous breed, were generally situated within the reference intervals previously reported for sheep.


Os valores hematológicos e bioquímicos, incluindo os eletrólitos plasmáticos e os hormônios da tireoide, foram determinados em 73 ovelhas, clinicamente saudáveis, da raça Churra da Terra Quente, raça ovina característica do nordeste de Portugal. Os valores obtidos para o hemograma foram: eritrócitos 9,8±1,5×10(12) /L; hemoglobina 118,1±19,1g/L; hematócrito 40,8±5,9 por cento; leucócitos 5,7±1,8×10(9) /L e plaquetas 544,3±177,2×10(9)/L. O tempo de trombina foi de 17,3±1,7 segundos. Os valores dos parâmetros bioquímicos avaliados foram: proteínas totais 76,4±6,1g/L; glicose 2,87±0,60mmol/L; colesterol total 1,65±0,33mmol/L; aspartato amino transferase 155,9±49,2U/L; alanina amino transferase 23,2±9,6U/L; gama-glutamil transferase 48,0±18,7U/L; fosfatase alcalina total 121,6±76,1U/L; glutamato desidrogenase 6,4±3,7U/L; ureia 7,32±2,22mmol/L; creatinina 123,0±54,1μmol/L; cálcio total 2,53±0,25mmol/L; fósforo 2,10±0,46mmol/L e magnésio 1,01±0,09mmol/L; sódio 152,04±3,65mmol/L; potássio 4,7±0,4mmol/L e cálcio ionizado 1,32±0,07mmol/L; tiroxina total 111,75±42,29nmol/L; tri-iodotironina total 1,01±0,28nmol/L; T4 livre 11,93±1,78pmol/L; T3 livre 4,22±1,33pmol/L e hormônio estimulante da tireoide 0,18±0,19μIU/mL. Apesar de terem sido observadas algumas diferenças entre a raça Churra da Terra Quente e outras raças, os valores hematológicos e bioquímicos, eletrólitos plasmáticos e hormônios da tireóide, desta raça autóctone apresentam-se no geral situados dentro dos intervalos de referência publicados para a espécie ovina.

ABSTRACT

OBJETIVOS: Determinar a frequência do fator reumatoide (FR-IgM), anticorpos antipeptídeos citrulinados cíclicos (anti-CCP), antinucleares (AAN), anticitoplasma de neutrófilos (ANCA), anticardiolipina (aCL) e anti-β2 glicoproteína I (anti-β2GPI) em pacientes com hanseníase, com e sem comprometimento articular, avaliando a possível associação entre estes autoanticorpos e as manifestações articulares, a forma clínica, a reação hansênica, o tratamento com poliquimioterapia (PQT) e a alta. PACIENTES E MÉTODOS: 158 pacientes com hanseníase foram distribuídos em dois grupos; 73 pacientes com (Grupo I) e 82 sem comprometimento articular (Grupo II). Compuseram o Grupo III 129 indivíduos saudáveis. MÉTODOS: aglutinação com partículas de látex para FR-IgM, imunofluorescência indireta para AAN e ANCA, e, ELISA para anti-CCP, aCL e anti-β2GPI. RESULTADOS: Dentre 158 pacientes com hanseníase, 56 apresentavam a forma virchowiana (VV). A frequência de anticorpos anti-CCP, FR e AAN nos Grupos I e II foi semelhante à do Grupo III. ANCA não foi detectado em nenhum dos grupos. Anticorpos aCL foram mais frequentes nos pacientes com hanseníase (Grupos I e II) que em controles sadios (15,8 por cento vs. 3,1 por cento; P < 0,001), não sendo observada diferença entre Grupos I e II (P = 0,67). Anticorpos anti-β2GPI também foram mais frequentes nos pacientes que nos controles (46,2 por cento vs. 9,4; P < 0,001), sem diferença significativa entre os Grupos I e II. Houve predomínio do isotipo IgM com relação ao IgG tanto para aCL (88 por cento vs. 16 por cento, P = 0,001), quanto para anti-β2GPI (97,3 por cento vs. 12,3 por cento, P < 0,001). Nenhum paciente apresentou manifestações sugestivas de trombose vascular. CONCLUSÃO: A frequência de anticorpos aCL e anti-β2GPI foi significativamente maior nos pacientes com hanseníase que nos controles saudáveis. OBJECTIVE: Determine the frequency of rheumatoid factor (IgM-RF) and anti-cyclic citrullinated peptide antibodies (anti-CCP), antinuclear antibodies (ANA), antineutrophil cytoplasmic antibodies (ANCA), anticardiolipin antibodies (aCL), and anti-β2 glycoprotein I antibodies (anti-β2GPI) in leprosy patients, with and without joint involvement, and to evaluate the possible association among those antibodies and articular manifestations, clinical type, reactional episodes, polychemotherapic treatment (PCT), and discharge from PCT. PATIENTS AND METHODS: One hundred and fifty-eight leprosy patients were divided in two groups of 73 patients (Group I) and 82 patients (Group II). Group III was composed of 129 healthy individuals. Methods: Semi-quantitative latex agglutination test for IgM-RF, indirect immunofluorescence for ANA and ANCA, and ELISA for anti-CCP, aCL, and anti-β2GPI. RESULTS: Fifty-six (35.4 percent) of 158 leprosy patients had lepromatous leprosy (LL). The frequency of anti-CCP, RF, and ANA antibodies in Groups I and II was similar to that of Group III. Antineutrophil cytoplasmic antibodies were not detected in any patient. Anticardiolipin antibodies were more frequent in leprosy patients (Groups I and II) than in control group (15.8 percent vs. 3.1 percent; P < 0.001), and differences between Groups I and II (P = 0.67) were not observed. Anti-β2GPI antibodies were also more common in leprosy patients than in control group (46.2 percent vs. 9.4 percent; P < 0.001), without differences between Groups I and II. A predominance of IgM isotype over IgG isotype was observed both for aCL (88 percent vs. 16 percent; P = 0.001) and anti-β2GPI (97.3 percent vs. 12.3 percent; P < 0.001). Patients did not present manifestations suggestive of vascular thrombosis. ABSTRACT

The objective of the present research was to evaluate the usefulness of anti-cyclic citrullinated peptide (anti-CCP) antibodies and the IgM rheumatoid factor (IgM RF) test for the differential diagnosis of leprosy with articular involvement and rheumatoid arthritis (RA). Anti-CCP antibodies and IgM RF were measured in the sera of 158 leprosy patients (76 with and 82 without articular involvement), 69 RA patients and 89 healthy controls. Leprosy diagnosis was performed according to Ridley and Jopling classification criteria and clinical and demographic characteristics of leprosy patients were collected by a standard questionnaire. Leprosy patients with any concomitant rheumatic disease were excluded. Serum samples were obtained from all participants and frozen at _20°C. Measurement of anti-CCP antibodies and IgM RF were performed by ELISA, using a commercial second-generation kit, and the latex agglutination test, respectively. Anti-CCP antibodies and IgM RF were detected in low frequencies (2.6 and 1.3 percent, respectively) in leprosy patients and were not associated with articular involvement. Among healthy individuals both anti-CCP antibodies and IgM RF were each detected in 3.4 percent of the subjects. In contrast, in the RA group, anti-CCP antibodies were present in 81.2 percent and IgM RF in 62.3 percent. In the present study, both anti-CCP antibodies and IgM RF showed good positive predictive value for RA, helping to discriminate between RA and leprosy patients with articular involvement. However, anti-CCP antibodies were more specific for RA diagnosis in the population under study.

ABSTRACT

We report an uncommon anatomical variations of the left renal vein was found on dissected specimen in an elderly male cadaver: a retroaortic left renal vein. No other vascular anomalies were noted in this specimen. The anomaly result may be related to a particular pattern of left inferior vena cava. The abnormality have to be known for it may be undetected or be misleading in imaging. This anatomical curiosity should be kept in mind by clinicians and academics that may manipulate this anatomical area.


Relatamos una variación anatómica infrecuente de la vena renal izquierda, denominada vena renal retroaórtica, encontrada en un cadáver masculino disecado. No se observaron otras variaciones vasculares. Lavariación anatómica encontrada se puede relacionar con un patrón particular de la vena cava inferior izquierda. La variación debe ser conocida ya que puede pasar inadvertida o ser confundida en proyecciones de imágenes de la región. Los clínicos y académicos que actúen en la región, deben considerar esta eventual variación anatómica.

ABSTRACT

Plants from the genus Alternanthera are thought to possess antimicrobial and antiviral properties. In Brazilian folk medicine, the aqueous extract of A. tenella Colla is used for its anti-inflammatory activity. The present study investigated the immunomodulatory property of A. tenella extract by evaluating the antibody production in male albino Swiss mice weighing 20-25 g (10 per group). The animals received standard laboratory diet and water ad libitum. The effect of A. tenella extract (5 and 50 mg/kg, ip) was evaluated in mice immunized with sheep red blood cells (SRBC 10 percent, ip) as T-dependent antigen, or in mice stimulated with mitogens (10 æg, Escherichia coli lipopolysaccharide, LPS, ip). The same doses (5 and 50 mg/kg, ip) of A. tenella extract were also tested for antitumor activity, using the Ehrlich ascites carcinoma as model. The results showed that 50 mg/kg A. tenella extract ip significantly enhanced IgM (64 percent) and IgG2a (50 percent) antibody production in mice treated with LPS mitogen. The same dose had no effect on IgM-specific response, whereas the 5 mg/kg treatment caused a statiscally significant reduction of anti-SRBC IgM-specific antibodies (82 percent). The aqueous extract of A. tenella (50 mg/kg) increased the life span (from 16 ± 1 to 25 ± 1 days) and decreased the number of viable tumor cells (59 percent) in mice with Ehrlich ascites carcinoma. The present findings are significant for the development of alternative, inexpensive and perhaps even safer strategies for cancer treatment

ABSTRACT

Despite some positive experiences, surgical rehabilitation, presently, is minimally available in endemic countries and the contingent of patients in need of this modality of treatment is enlarging constantly. With the reduction of prevalence of leprosy and the progressive integration of care of leprosy-affected persons into the general health services, surgical rehabilitation should be made available in these centres. Training health personnel of general health services in leprosy surgery is mandatory and urgent. There is a need for the settlement of a multi-organizational task-force (WHO-ILEP-ILA) to deal with this problem. The aim of this task-force should be the definition of strategies to organize such training and to increase awareness among medical societies and schools regarding this issue. This group should also remember that the referral centres in Africa, Asia, North and South America should act as reservoir of this knowledge and, as such, should be fully supported and enhanced, since they have an important and unique role to play in the training of general health services personnel.

ABSTRACT

Objetivo. Estudar as normas relativas à ética da pesquisa em seres humanos contidas nas instruções aos Autores de revistas científicas brasileiras. Material e Métodos. As Instruções aos Autores de 139 revistas científicas brasileiras das áreas de medicina, biomedicina, enfermagem, odontologia e ciências gerais foram analisadas com relação às suas recomendações sobre os aspectos éticos. Resultados. Das 139 revistas estudadas, 110 (79,1 por cento) não fazem referências aos aspectos éticos; 17 (12,2 por cento) exigem aprovação prévia pela Comissão de Ética; três (2,1 por cento) fazem referência à Declaração de Helsinque; uma (0,7 por cento) recomenda adotar o consentimento esclarecido; cinco (3,5 por cento) seguem orientações dos requisitos uniformes para manuscritos submetidos a revistas biomédicas e três (2,1 por cento) seguem princípios, normas e padrões éticos não especificados. Nas 29 revistas que fazem referências à ética, as exigências são solicitadas sob as seguintes formas: 15 (51,7 por cento) exigem a inclusão da informação no texto do artigo; duas (6,8 por cento) pedem carta assinada pelos autores; uma (3,4 por cento) solicita cópia da autorização da Comissão de Ética; uma (3,4 por cento) afirma supor que o autor cumpriu as exigências e dez (34,4 por cento) não fazem qualquer referência específica. Conclusões. Os resultados do estudo indicam que na maioria das revistas científicas brasileiras há pouca preocupação em relação aos aspectos éticos da pesquisa em seres humanos, contidas nas Instruções aos Autores. Nas revistas que fazem referência aos aspectos éticos, as exigências são muito variáveis.

Novos ribonucleosídeos derivados dos sistemas dipirazolo-piridina foram preparados e avaliados quanto à atividade polimerásica das enzimas transcriptase reversa (RT) do vírus HIV-1 e das DNA polimerases humanas alfa e epsilon. Os derivados 1b e 1d inibiram a atividade da transcriptase reversa em concentraçöes de micromolares. Entretanto, as mesmas substâncias näo foram capazes de inibir a atividade polimerase das enzimas DNA-polimerase humana alfa e epsilon.

Pacientes de una zona predominantemente deportiva fueron evaluados mediante ultrasonidos, un examen rápido, inocuo y dinámico de músculos y tendones. Se efectuó el estudio de las regiones aquejadas comparativamente en relajación y con maniobras de contracción, dorsiflexión, abducción y aducción. Los estudios, principalmente de recto anterior, cuádriceps, isquiobiales, gemelar interno bíceps y tendón de Aquiles permitieron en general inferir el estado y gravedad de las lesiones

ABSTRACT

Squamous cell carcinoma of the esophagus has a poor prognosis. There are several factors influencing the prognosis. The study of DNA ploidy by flow cytometry used in many malignant tumors is very valuable in determining, as a tumor marker, this prognosis. In this study 13 cases of resected squamous cell carcinoma of the esophagus were analyzed. Despite the small sample, the authors observed that the worst prognosis was in advanced stage cases (III and IV), an aneuploid DNA pattern. The only case with a 5-year survival had a diploid DNA pattern.

ABSTRACT

Stimulant properties during exercise have been attributed to cafeine (CAF) and tryptophan (Trp). The purpose of the present study was to investigate the effects of CAF and Trp ingestion on rectal temperature (Tre), total exercise time (TET), oxygen consumption (VO2), carbon dioxide production (VCO2) pulmunary ventilation (VE), heart rate (HR) and rate of perceived exertion (RPE) during exercise on a cycle ergometer at 80 percent of maximal work load, in eight halthy male volunteers. Each subject abstained from caffeine for 48 h and from animal-derived foods for 36 h before each experiment. Aerobic capacity was determined on the first day. In consecutive trials, conducted in a double-blind, randomized, crossed-over manner, each subject recived capsules containing CAF (10mg/Kg), Trp (1.2g), a combination of the two (CAF + TRP), and lactose (PLA), 1 h before exercise. Plasma CAF concentration (PC) was measured by high performance liquid chromatography (HPLC), before (basal concentration) and 1 and 2 h after ingestion of the capsules. At both times after CAF or CAF + Trp ingestion, the PC was elevated compared with the basal concentration (P < 0.05). During exercise, significant increases occured with time in Tre, TET, VO2, VCO2, VE, HR and RPE (P < 0.01) while no significant difference was observed when CAF or CAF+ Trp were compared with control values. Under the conditions of this study, CAF and/or Trp did not affect the physiological parameters measured before, during or after exercise at 80 percent of maximal work load

We present a case of gastric metastasis, leading to chronic bleeding in a patient already submitted to surgery for a malignant melanoma of the lip. In view of the good clinical conditions, a subtotal gastrectomy as palliative treatment was performed. We discuss the diagnostic and therapeutic aspects of this rare clinical situation

Se evaluaron 30 pacientes con enfermedades reumáticas (artritis reumatoidea, LES, colagenopatías mixtas) y diversas afecciones articulares con preponderancia del estudio ultrasonográfico sobre patología bursal. Determinando la ubicación, extensión, forma y contenido o presencia de derrame articular asociado, definimos patrones ecográficos de las bursitis del hombro, codo, rodilla, tobillo y cadera (trocanteritis mayor), bases sonográficas para el seguimiento evolutivo de esta frecuente afectación reumatológica

ABSTRACT

Se estudiaron 90 pacientes con supuesta patología de las glándulas salivales mediante sialografía y ecografía. El objetivo del estudio fue establecer la capacidad diagnóstica de cada método en particular, en distintas patologías: litiasis extra e intraglandular, afecciones inflamatorias de los conductos excretores y de las glándulas y tumores. Se determinó además la incidencia de la patología por sexo y grupo etario. Se establecieron las indicaciones de cada método cuya eficiencia es semejante en algunos casos y absolutamente complementaria en otros. Ambos no pueden reemplazarse entre sí ni excluyen otros medios de diagnóstico por imágenes a los que se deberá apelar en circunstancias definidas como la TC y la RNM

ABSTRACT

Evidência sorológica de infecçäo por rotavírus em uma colônia de cobaios - Anticorpos reagindo com rotavírus símio SA11 foram demonstrados por ensaio imuno-enzimático (EIE) e por "Western blot assay" (WBA) em soros de cobaios mantidos para fins experimentais na Fundaçäo Oswaldo Cruz, Rio de Janeiro, Brasil. A proporçäo de animais soro-positivos e os níveis de anticorpos subiram rapidametne em 1985, mantiveram-se altos em 1986 e baixaram em 1987. Näo foram observados sinais de doença coincidente com a elevaçäo de anticorpos. Resultados de WBA sugerem que o rotavírus responsável pela resposta sorológica pertence ao subgrupo do grupo A

Isolamento e caracterizaçäo antigênica do vírus da imunodeficiência humana (VIH/HIV) no Brasil - Um retrovírus foi isolado de um paciente brasileiro com "Síndrome de Imunodeficiência Adquirida" (SIDA/AIDS) e caracterizado em termos de sua reatividade com soros de indivíduos infectados com vírus da imunodeficiência humana dos tipos 1 e 2 (HIV-1 e HIV-2). A análise antigênica por "Western blot" revelou que o isolado brasileiro é bastante similar a uma cepa de HIV-1 bem caracterizada. A identificaçäo do retrovírus como HIV-1 e näo HIV-2 é reforçada pelo fato dos anticorpos do paciente do qual foi isolado o vírus näo terem reagido com a glicoproteínas de envelope de 140 kDa, específica para HIV-2

ABSTRACT

Foram estudados soros de pessoas de diferentes grupos etários coletados em 1976, 1978 e 1979 para verificar a presença de anticorpos inibidores da hemaglutinaçäo contra diversas estirpes de vírus da influenza A dos subtipos H3N2 e H1N1. A ocorrência da infecçäo pelo subtipo H3N2 foram detectados em 1976 e 1978 mas em 1979, a circulaçäo desse subtipo de vírus foi limitada. A prevalência de anticorpo contra A/Säo Paulo/1/78 (H1N1) foi significativamente maior do que para A/USSR/90/77 (H1N1) em 1978. No entanto em 1979, a estirpe predominante foi A/USSR/90/77 (H1N1). As pessoas com idade inferior a 20 anos foram as mais afetadas pelo subtipo H1N1, enquanto que indivíduos com mais de 20 anos já apresentavam anticorpos para esse subtipo em 1976, antes do resurgimento dessa estirpe. A infecçäo pelo vírus H3N2 em 1978 ocorreu em 65,4% de crianças do grupo etário de 0-4 anos; 47,0% de criancas do mesmo grupo tinham anticorpos para vírus H1N1 em 1979. Anticorpos para o vírus da influenza suina foram detectadas em 60% de pessoas com mais de 60 anos de idade
